Voici la présentation de la petite expérience que nous allons effectuer au labo la semaine prochaine
** Protocol**
For this experiment we will need the following :
1) a petri dish containing agar solution.
2) A sample of yeast in a glass bottle
3) An inoculating loop
4) A Bunsen burner
5) matches
Time required: less than 5 minutes to streak; 2-3 days for the colonies to grow
Procedure
We must start by turning on the gas & using a match to light the Bunsen burner.
Then we open the top of the glass bottle containing the sample of yeast with our little finger or pinky finger. At this stage we must sterilize the top of the bottle by passing it over the flame of the Bunsen burner.
Now we take the oculating loop and sterilize it by holding the tip over the Bunsen burner for a couple of seconds before letting it cool down. If we forget to let it cool we will kill the yeast.
Now we use the loop to take a sample of yeast from the bottle before sterilizing the top of the bottle again & putting the lid back on it.
At this stage we have to take the lid off our petri dish containing the agar solution.
To transfer the cells to the media plate,we must begin the streak at one edge of the plate. We must press the side of the loop containing the cells to the agar plate’s surface and quickly streak back and forth across part of the plate’s surface. The streaks should lie near one another, but should not cross over previous streaks.
When we have finished streaking out the first side of the plate, we must turn the plate around a quarter of a turn before streaking out the second side of the plate.
At this stage we must re-sterilize the inoculating loop by running it through the flame of the Bunsen burner & letting it cool down to avoid killing the yeast.
We start to streak out the second side of the plate. These streaks should start by crossing over the last streak we made, then proceeding as before into new areas of the agar plate.
After this we must turn the plate around a quarter of a turn again before streaking out the last side.
We must sterilize our inoculating loop again & let it cool down.
To streak out the last side, we have to do spread the yeast on the last side of the agar plate by making the same back & forth movements with the inoculating loop. The cells will be distributed on the plate in decreasing concentration through the streaks, and no matter how many cells were on the inoculating loop to begin with, there should be an area on the streaked plate that will produce isolated colonies.
Cover the plate, invert it and incubate at 37˚C for 2-3 days. The plate should appear to have solid streaks of cells as well as isolated colonies.
Check the plate after 2 to 3 days’ growth.
